Promoter of Agrobacterium VirD2-Binding Protein
Usage and Biology
The Pvbp2 promoter is an inducible promoter for VirD2-Binding Protein originated from Agrobacterium tumefaciens str.C58 The Pvbp2 promoter was cloned into reporter plasmids containing Green Fluorescent Protein (GFPuv, BBa_M45116) and transformed into Agrobacterium tumefaciens GV3101 for further testing.
Acetosyringone (AS) is the most commonly used inducer for transgene induction in Agrobacterium tumefaciens, and an important function of VBP proteins is to recruit T-complexes to type IV secretion systems[1]. So we tried to induce the expression of this promoter using acetosyringone. According to relevant literature, the concentration of the inducer was about 150μM[2].
An examination of chromaticity values, obtained at consistent exposure times using a multi-function imager, reveals significant disparities. The basal fluorescence of the culture was subtracted in this analysis.
